چکیده
www.landesbioscience.com Cell Cycle 3427 The molecular details of how lamin A influences 53BP1 stability and inhibits formation of p130/e2F4 repressor complexes remain to be identified. Lamins may directly bind to and affect these proteins, or these effects may be indirect. Altogether, the study by Redwood et al. provides a glimpse of novel functions of lamin A in both the transcription and stabilization of DnA damage repair components. Lamin A thus seems to also be involved in regulating the intricate cross talk between the different DsB repair pathways. References 1. stracker TH, et al. nat Rev Mol Cell Biol 2011; 12:90-103. 2. Spinophilin in human cancer-molecular mechanisms and clinical relevance
Spinophilin, a multifunctional intracellular protein, has attracted attention within the last years as a novel putative tumor suppressor protein. Recent studies have shown a reduction of spinophilin expression levels in various types of cancer, such as lung adenocarcinoma, ovarian cancer, and chronic myelogenous leukemia. Low expression of spinophilin was also associated with a higher malignant...
متن کاملSpinophilin expression determines cellular growth, cancer stemness and 5-flourouracil resistance in colorectal cancer
The putative tumor suppressor gene spinophilin has been involved in cancer progression in several types of cancer. In this study, we explored the prognostic value of spinophilin expression in 162 colon adenocarcinoma patients. In addition, we generated stably expressing spinophilin-directed shRNA CRC cell lines and studied the influence of spinophilin expression on cellular phenotypes and molec...
متن کاملLow spinophilin expression enhances aggressive biological behavior of breast cancer
Spinophilin, a putative tumor suppressor gene, has been shown to be involved in the pathogenesis of certain types of cancer, but its role has never been systematically explored in breast cancer. In this study, we determined for the first time the expression pattern of spinophilin in human breast cancer molecular subtypes (n = 489) and correlated it with survival (n = 921). We stably reduced spi...
متن کاملThe Role of Tumor Protein 53 Mutations in Common Human Cancers and Targeting the Murine Double Minute 2–P53 Interaction for Cancer Therapy
The gene TP53 (also known as protein 53 or tumor protein 53), encoding transcription factor P53, is mutated or deleted in half of human cancers, demonstrating the crucial role of P53 in tumor suppression. There are reports of nearly 250 independent germ line TP53 mutations in over 100 publications. The P53 protein has the structure of a transcription factor and, is made up of several domains. T...
متن کاملDeep Insight Section
Spinophilin is a scaffolding protein with modular domains that govern its interaction with a large number of cellular proteins. The Spinophilin gene locus is localized at chromosome 17q21, a chromosomal region frequently affected by genomic instability in different human tumours. The scaffolding protein interacts with the tumour-suppressor ARF which has suggested a role for Spinophilin in cell ...
